Water over the front edge
In a hard downpour a full gutter sends rain straight past the channel and down your siding instead of through the downspout.

Something growing in the channel
Maple seeds and pine needles make a fine seed bed. If plants are sprouting up there, water is not moving through.

Streaks down the gutter face
Overflow stains the front of the gutter and the wood behind it, and it sets in harder the longer it sits.

A sagging or separated run
Full gutters are heavy. Hangers pull, joints open, and the water starts finding the gaps instead of the downspout.
